چکیده
vaccines against the hiv-1 virus offers the best hope for eliminating hiv-associated mortality. recombinant adenovector type 5 (rad5) vaccine is a potential candidate for preventive vaccine strategies. in this study, we evaluated the rad5 prime/protein boost strategy in a murine model. we used rad5 harboring single hiv-1 genes. these genes, including gag (p24) and exon1 of tat, were amplified from hiv-1 (clade a) rna using nested pcr. recombinant vectors were constructed, purified and then injected at 10 12 viral particles into four groups, each comprising five mice. the groups were each assigned to receive one of rad5 prime/protein boost gag, tat with and without recombinant granulocyte-macrophage colony-stimulating factor (rgm-csf), and rad5 with and without genes. the humoral responses were evaluated using elisa and cellular immune responses checked by cell proliferation and elispot assays (il-2, il-4 and ifn-γ). it was shown that compared with the rad5 injection alone, the rad5 prime/protein boost plan increased cellular immunity ( p = 0.009) as well as humoral immunity ( p = 0.009). moreover, rgm-csf as an adjuvant enhanced cell-mediated immunity and increased il-4 expression ( p =0.032). the results revealed that the simultaneous use of multiple antigens and heterologous prime/boost strategy can enhance both humoral and cellular immune systems. moreover, subcutaneous injection of rgm-csf increases il-4 production and shifts the immune pattern to th2 . these strategies can potentially be used to develop an efficient hiv-1 vaccine.

متن کاملCharacterization of Immune Responses Induced by Combined Clade-A HIV-1 Recombinant Adenovectors in Mice
Background: Numerous evidences indicate that in some HIV-1 positive patients, the humoral and cellular immune responses are induced against HIV-1 proteins and this is inversely related to the progress of infection. Objective: The aim of this study was the evaluation of the Adenovectors containing HIV genes in induction of immune responses in mice. Methods: The HIV-1 genes including gag p24, rev...
متن کاملIL-12 and GM-CSF in DNA/MVA Immunizations against HIV-1 CRF12_BF Nef Induced T-Cell Responses With an Enhanced Magnitude, Breadth and Quality
In Argentina, the HIV epidemic is characterized by the co-circulation of subtype B and BF recombinant viral variants. Nef is an HIV protein highly variable among subtypes, making it a good tool to study the impact of HIV variability in the vaccine design setting. We have previously reported a specific cellular response against NefBF with low cross-reactivity to NefB in mice. The aim of this wor...
متن کاملDNA prime and protein boost immunization with innovative polymeric cationic core-shell nanoparticles elicits broad immune responses and strongly enhance cellular responses of HIV-1 tat DNA vaccination.
Novel biocompatible core-shell cationic nanoparticles, composed of an inner hard core of poly(methylmethacrylate) (PMMA) and a hydrophilic tentacular shell bearing positively charged groups and poly(ethyleneglycol) chains covalently bound to the core, were prepared by emulsion polymerization and characterized in vitro and in vivo for DNA vaccine applications. The nanoparticles reversibly adsorb...
متن کاملGM-CSF Fails to Improve Immune Responses to Booster Hepatitis B Vaccination in HIV-Infected Individuals
BACKGROUND Hepatitis B (HBV) vaccination is an important preventive intervention for HIV-infected population. Data regarding booster HBV vaccine for persons with low HBV surface antibody (sAb) titers after vaccination in this immunocompromised population is lacking. METHODS We randomized 60 HIV-infected subjects lacking HBV protection after completion of 3 doses of HBV vaccine to receive a bo...
متن کاملA Novel Prime and Boost Regimen of HIV Virus-Like Particles with TLR4 Adjuvant MPLA Induces Th1 Oriented Immune Responses against HIV
HIV virus-like particles (VLPs) present the HIV envelope protein in its native conformation, providing an ideal vaccine antigen. To enhance the immunogenicity of the VLP vaccine, we sought to improve upon two components; the route of administration and the additional adjuvant. Using HIV VLPs, we evaluated sub-cheek as a novel route of vaccine administration when combined with other conventional...
